Overview
In this viability assay, the total number of nucleated cells and the total number of dead cells are identified by staining the cells with Hoechst and Propidium Iodide (PI). Propidium Iodide is membrane impermeable and only enters the dead (membrane compromised) cells, where upon entry it binds to DNA in an intercalating manner. Hoechst 33342 is a DNA binding agent that is membrane permeable and is used to identify all nucleated cells.
