Transform cell line development (CLD) and manufacturing of biologics with CHOSOURCE™ TnT transposon technology. The CHOSOURCE TnT redefines the CLD paradigm and can help accelerate biologic development programs.

CHOSOURCE TnT comprises two elements: the TnT transposon vector and the TnT transposase provided as messenger RNA (mRNA).

The TnT transposon vector, where the gene of interest (GOI) is cloned and integrates, contains:

  • Dual expression cassette for multi-chain protein expression
  • An optimized GS selection cassette for enhanced productivity, with no MSX addition required
  • A precisely engineered Terminal Inverted Repeats (TIRs) flanking the expression cassettes for efficient genome integration.

How does CHOSOURCE TnT transposon technology work?

The TnT transposase, co-transfected with the TnT vector in the host cell, mediates the integration of the expression cassettes from the TnT vector (donor) into transcriptionally active regions of the host genome. This controlled DNA delivery helps enable highly efficient and stable integration of the GOI into the host genome, as well as assists in increasing cell line expression performance and clone stability while reducing uncertainty and variability during cell line development. Selecting high-expressing clones becomes effortless.

Faster recovery time during pool selection and high reproducibility across pools and clones can help accelerate cell line development and the manufacturing process.

Safe and reliable genetic modification of host cells

The TnT transposase is delivered as mRNA to help reduce the risk of integration into the host genome. Transposase enzyme and transposase mRNA are degraded shortly after transfection, avoiding generation of genomic alterations and/or instabilities.

The consistent high transfection efficiency and clonal stability achieved with CHOSOURCE TnT significantly reduces unexpected setbacks during cell line and process development.

Streamline process development

CHOSOURCE TnT assists users with effortlessly generating stable expressing clones without the use of MSX or any other selection agent. The high reproducibility observed at pool and clone levels, reduces the efforts required for screening many pools and/or clones, thus simplifying the CLD process.

Disruptive development and manufacturing workflows

The robustness and reliability of CHOSOURCE TnT makes it a great tool to support streamline development and manufacturing efforts, potentially allowing new development paradigms to expedite preclinical (and potentially clinical) development.

CHOSOURCE TnT and CHOSOURCE GS KO and/or ADCC+ cell lines are available in one combined royalty-free license, providing a simple technology access framework without the need for any other third-party technology license. Available for commercial and research applications.

The CHOSOURCE Platform is available for research, clinical, diagnostic, and commercialization applications under specific licenses from Revvity. The platform is also available for services under a service license from Revvity.
